Back before I had a kid, I used to take weekly Pilates classes at a local studio. While many of the moves bore passing resemblance to those we do in my yoga class, it was unabashedly a workout?a sweat-inducing, core-honing, fast-paced workout. I loved it (and so did my abs). But then my daughter came along and something had to give, so I said goodbye to Pilates, but I missed it!

So when I had the opportunity recently to test out two home Pilates systems, I was delighted! Here?s what I discovered:

Pros: This deceptively simple system (it?s just a bar with a resistance cord and foot strap at each end) is easy to put together, and you can follow the workouts on the DVD or just go free-style, twisting, curling, and lifting in all sorts of directions. The DVD is versatile, offering three workouts (plus a bonus arms sequence) that you can mix and match. Instructor Gin Miller gives excellent direction, and while the routines are pretty simple, they?re still challenging.
Cons: The quick-reference poster only offers six exercises.
Cool extra: The moves are often shown in split screen, from two different angles?really helpful when it comes to getting just the right form.

Pros: Where do I start? I loved this! It?s sturdy and easy to assemble, and the end bar unscrews to store inside the body of the system (called the Roller) when you?re done. The cords have three levels of resistance, and you can switch from one to the other with, literally, the press of a button. I can?t say enough about the accompanying DVD?trainer Jennifer Galardi gives terrific, detailed instructions (she?s very likeable), and the workouts combine strength, cardio, balance, and lots of core work for a true full-body challenge. But don?t get me wrong?you don?t have to use the DVD. My daughter and I had a blast using it as a balance beam, then making up our own moves with the resistance cords. More good stuff: It also comes with a track-your-workouts poster, a collection of workout cards, and a washable cover.
Cons: There?s a carrying strap so you can schlep the Reformer around, but it?s pretty heavy, so you may not want to go far. Also, the two Stabilizers that hold the Roller in place don?t fit inside the unit for storage.
Cool extra: Use it like a foam roller for an awesome massage. You can either follow the ?Renew? part of the Stretch & Renew routine, or just do whatever feels good on your deserving muscles.

Researchers identify mechanism that makes breast cancer invasive

A new study has identified a key mechanism that causes breast cancer to spread. The research, published by Cell Press on March 30th in the journal Molecular Cell, enhances our knowledge about the signals that drive cancer metastasis and identifies new therapeutic targets for a lethal form of invasive breast cancer that is notoriously resistant to treatment.

Amplification of the gene for ErbB2 has been linked with aggressive forms of breast cancer and is associated with a poor outcome. Although ErbB2-positive breast cancers are routinely treated with chemotherapy and the ErbB2-targeted drug trastuzumab (Herceptin), this treatment often fails. "Nearly half of breast cancers with high ErbB2 expression are either initially non-responsive or eventually develop resistance to trastuzumab," explains senior study author, Dr. Tuula Kallunki, from the Danish Cancer Society Research Center. "Therefore, a better understanding of the molecular basis of ErbB2-induced malignancy is critical for further development of targeted treatments."

In the current study, Dr. Kallunki and colleagues discovered that ErbB2 induces expression of proteins called cathepsins. Previous work has linked cathepsins with metastasis of pancreatic cancer, specifically with tumor cell invasion of surrounding tissues and with the establishment of tumor blood supply. Using a three-dimensional model system to study ErbB2-induced cellular invasion, the researchers demonstrated that inhibition of cathepsins prevented ErbB2-driven breast cancer invasion. They went on to identify components of the complex signaling network linking ErbB2 and cathepsin expression with cancer invasiveness.

Taken together, the results demonstrate that the invasive behavior of ErbB2-positive cancer cells is dependent on cathepsin activity and uncover a molecular link between ErbB2 and breast cancer invasion. "Our work has introduced a cellular model system to study ErbB2-induced invasion and identified an invasion-promoting signaling network," concludes Dr. Kallunki. "In addition, our findings provide several potential therapeutic targets against ErbB2-driven invasive cancers."

Genome study confirms immune system link to disfiguring leg swelling

Genetic variants in a region of the genome linked to our immune response have been linked to increased risk of podoconiosis, a disfiguring and disabling leg swelling caused by an abnormal reaction to the minerals found in soil. An estimated 4 million people worldwide suffer from the condition.

In a study published today in the New England Journal of Medicine, researchers funded by the Wellcome Trust and the Association of Physicians of Great Britain and Ireland compared the genomes of 194 people affected by the disease from southern Ethiopia against 203 people who were unaffected. They identified three genetic variants that increased the risk of developing the condition.

Podoconiosis, or 'podo', as it is often called, was added to the World Health Organization's list of neglected tropical diseases in 2011. It is a type of elephantiasis (leg swelling) found in farming communities in the tropics and is triggered by an abnormal reaction to irritant mineral particles found in soils of volcanic origins amongst people who cannot afford shoes.

Many years of walking, ploughing or playing barefoot on these soils appears to trigger inflammatory changes within the lymph system in the legs, which in time can lead to foot swelling and ultimately elephantiasis.

The disease often runs in families, implying that there is a hereditary component to the disease, but until now, no genetic variants had been identified which confer increased risk. The genetic variants discovered in this new study all fall within a region of the genome known as the HLA class II, which is important in controlling immune responses. Combined, the three variants increase the risk of developing podoconiosis by a factor of two to three.

Prof Melanie Newport from Brighton & Sussex Medical School, who led the study, says: "The region where we have found these susceptibility genes for podoconiosis plays an important role in controlling our immune system. It confirms what we had expected, that there is an immunological basis to the disease. Although this is still early days for identifying potential treatments, it suggests that drugs that target immune responses may be useful."

First author Dr Fasil Tekola Ayele from the Armauer Hansen Research Institute, Ethiopia, adds: "Genome wide association studies on African populations are still fairly novel. However, this study highlights the importance of such studies in helping us understand the origins of diseases that are particularly common on the continent." Dr Ayele is currently on a postdoctoral attachment at the National Human Genome Research Institute, USA.

Dr Abraham Aseffa, also from the Armauer Hansen Research Institute, says: "Our next step is to try to pinpoint exactly which molecules are involved in podoconiosis, and which specific genetic mutations affect the function of these molecules. This will shed a lot more light on potential therapeutic options."

Professor Newport and colleagues have recently launched Footwork, an international initiative to bring together public and private partners to prevent and treat podoconiosis Footwork aims to integrate podoconiosis control with that of other neglected tropical diseases wherever possible, and to partner with organizations working in foot-related conditions to advocate for shoes as cost-effective interventions to tackle such diseases.

"There are still many places round the world where people cannot afford a pair of shoes," says Dr Gail Davey, co-author on the study and Executive Director of Footwork. "For some people, this means cold, cut or bruised feet, but for others it can lead to podoconiosis, which can have a significant impact on their quality of life. We hope that shoes can become the 'new bed-nets': simple, cost-effective interventions that mean that in future there is no reason for anyone's life to be destroyed for the want of a shoe."

Commenting on the findings, Dr Jimmy Whitworth, Head of International Activities at the Wellcome Trust, adds: "Podoconiosis is finally getting the attention it deserves as a disease that blights the lives of millions of people. The success of this international collaboration is testament to the importance of providing opportunities for training and building capacity for African researchers to take a lead on important work such as this."

Microbial Mules: Scientists Experiment With Engineering Bacteria to Transport Nanoparticles and Drugs

Bacteria are exquisite living machines that could one day deliver medicine to cells in the human body, if scientists overcome the numerous obstacles

Tiny robots that swim through our blood vessels attacking viruses and malignant cells have not quite crossed the line that separates science fiction from science?but there might be a way to jump-start their development.

Engineering nimble robots that are smaller than blood cells is extremely challenging. Rather than design them from scratch, some scientists have been experimenting with the idea of enlisting an army of sophisticated nanobots already at our disposal: the thousands of species of bacteria swarming inside our bodies right now. In recent years researchers have saddled microorganisms with useful nanoparticles and bits of DNA. Although the research is preliminary, some engineers and microbiologists see realizable potential. This week, at the American Chemical Society's annual meeting in San Diego, biomolecular engineer David Gracias of Johns Hopkins University discussed the progress he and his colleagues have made in gluing nanoparticles to bacteria.

So far, Gracias and his colleagues have managed to decorate Escherichia coli with incredibly tiny beads, rods and crescents made from nickel and tin coated in gold. Each nanoparticle is about 300 nanometers in diamter, or three times the size of a particle of wood smoke. The researchers stuck the nanoparticles to the bacteria with antibodies, which are small, Y-shaped proteins that the immune system uses to identify foreign cells, viruses and harmful bacteria. Each antibody fits like a puzzle piece into an array of different proteins on the surfaces of invading pathogens; antibodies can also lock onto other antibodies?usually from a different species. Gracias and his colleagues first coated the nanoparticles with antibodies from goats and rabbits, and subsequently coated the bacteria in complementary antibodies. When they soaked the bacteria in a solution of the nanoparticles and heated the mixture, the beads, rods and crescents stuck to the bacteria, like so many Lucky Charms marshmallows clinging to the underside of a wet spoon.

Gracias and his teammates also devised a way to load bacteria with nanoparticles at tiny docking stations and release them on demand. First they covered small squares of silicon and gold with a field of Y-shaped antibodies, to which they attached small beads dotted with complementary antibodies. Then they introduced bacteria sporting another set of complementary antibodies to the awaiting beads?as expected, they clasped like Velcro. A wave of dissolving chemicals detached the bacteria and their cargo beads from the docking station.

Many bacteria saddled with nanoparticles were still able to move around freely, albeit not as quickly as their unencumbered brethren. Sometimes, though, the backpacking bacteria just spun in circles, apparently unable to move forward. "This is definitely a work in progress," Gracias says. "Right now we are mostly focusing on varying the size and shape of the nanoparticles and making sure they stick."

Such nanoparticles can be heated from afar with infrared light, destroying diseased tissue. Ultimately, Gracias dreams of coaxing bacteria to ferry spongy nanoparticles soaked in drugs and outfitting bacteria with tiny sensors that measure local temperature and pH or tiny tools that perform surgery on a single cell. Similar research by other scientists confirms the potential of engineered bacteria to deliver medical packages directly into living cells.

In earlier work, Demir Akin of Stanford University and his colleagues used antibodies and nanoparticles to attach molecules of DNA to weakened Listeria monocytogenes, which is a bacterium responsible for many cases of food poisoning. L. monocytogenes is an intracellular bacterium, which means it has evolved ways to get inside animal cells. Akin attached a luciferase gene?which codes for the enzyme that makes fireflies glow?to L. monocytogenes and injected the germs into living mice. Three days later the mice glowed under a specialized camera, confirming that not only had the bacteria entered the mouse's cells, the cell nuclei had incorporated the bacteria's cargo and expressed the gene. Akin designed the living microbots to release their DNA packages inside mammalian cells, where the pH is low (acidic) enough to dissolve the chemical bonds gluing the luciferase gene to the bacteria.

The advantage of L. monocytogenes is that it knows how to get into cells, but it would be risky to use even a weakened version as a medical workhorse because it makes people sick. E. coli, in contrast, are much more harmless, but not all strains have specific adaptations for entering cells. The key, says Douglas Weibel of the University of Wisconsin?Madison, is working with a harmless microorganism that is a strong swimmer and has no problem butting its way into mammalian cells. In an experiment they conducted largely for fun, Weibel, George Whitesides of Harvard University and their colleagues yoked nanosize polystyrene beads to a single-cell green alga called Chlamydomonas reinhardtii. Weibel and Whitesides successfully steered their "microoxen" by shining light on one side of the algal cells (algae move toward light).

After that, Weibel did not pursue the challenge of engineering microorganisms to ferry nanoparticles, but he remains fascinated by the ongoing research. "Bacteria have already figured out how to move around in the body," he says. "They have evolved amazing motility. They can sense changes in their environment and adapt, not only on a short timescale, but they can adapt genetically, too. Even if we can't get them to deliver things in the human body, they could be useful for transporting nanoparticles in the lab. Who knows what advances we'll have 50 years from now."
